Is it possible to create an index or listing file with all available downloads and their description in it?
Would be very handy to see if a file is already posted and for users to browse it off-line.

You could use the List All Downloads menu item.  Attached is a pic using a compact layout. Here Compact means that the Info block is horizontal.  It is not 'narrow' like the others I am working on for jD v3.9
You can get compact layouts from http://www.jdownloads.com/index.php/downloads/category/11-files-layouts.html

I meant like exporting a listing to a file you you can read and print it if necessairy.
(it's easier if you can print it and 'walk through' very quickly...)

Another question:
Is there a way to always show the number of downloads in the header? (now they are only shown with 'all downloads')

There is no facility to produce a file or a printed report as you would like.  Could examine possibilities when we get jD3.9 released.  It would probably be a module.

In your second query you can use the jD Stats module.  This reports the number of downloads, categories and total Downloads
